Aeterna Zentaris to stop late-stage cancer drug trial, shares fall

(Reuters) - Canadian drugmaker Aeterna Zentaris Inc said it would discontinue a late-stage trial of a cancer drug after an independent data safety monitoring committee said the drug was unlikely to help increase patient survival.

Shares of the company fell as much as 29 percent to C$1.89 on Monday on the Toronto Stock Exchange.

The committee recommended that patient enrollment be stopped and the study discontinued for perifosine, the company said in a statement.

The drug is being studied to treat multiple myeloma, a cancer that starts in plasma cells in bone marrow and disrupts the production of normal blood cells.

The U.S. Food & Drug Administration granted perifosine an orphan-drug designation in September 2009 as well as a fast-track designation in December 2009.

The late-stage trial compared the efficacy and safety of perifosine to a placebo when combined with bortezomib and dexamethasone in patients with multiple myeloma.

The committee also said it was highly unlikely that the study would achieve a significant difference in its primary endpoint and progression free survival, or increasing survival of patients without the cancer worsening.

Widow's wish resurrects 'Mind the Gap' recording

LONDON (AP) ? A widow's wish to hear her late husband's voice again has prompted London Underground to restore a 40-year-old recording of the subway's famous "mind the gap" announcement.

The subway system ? better known as the Tube ? said it has tracked down the voice recording by Oswald Lawrence after his widow, Margaret McCollum, wrote to ask for a copy of it when she noticed it was not broadcast in the system anymore.

McCollum said she used to frequently visit Embankment station to hear her husband's voice, but was disappointed when it wasn't there last year.

Nigel Holness, director of London Underground, said its staff has been so moved by McCollum's story that they dug up the recording and gave the widow a copy of the announcement on a CD for her to keep. Tube staff is also working to restore the announcement at the station, he added.

McCollum said Sunday she has been overwhelmed by the media attention to her story.

"I had no idea this was going to happen ? I'm very touched by all the interest in it," she said.

The Tube's automated "mind the gap" messages, voiced by various actors, have accompanied countless London commuter journeys since the 1960s. Train drivers and staff made the warnings themselves before that.

London's subway, the world's first underground railway network, was begun in 1863. It is celebrating its 150th anniversary this year.

Antibiotics are unique assassins

In recent years, a body of pub?li?ca?tions in the micro?bi?ology field has chal?lenged all pre?vious knowl?edge of how antibi?otics kill bac?teria. "A slew of papers came out studying this phe?nom?enon, sug?gesting that there is a gen?eral mech?a?nism of killing by antibi?otics," said Kim Lewis, Uni?ver?sity Dis?tin?guished Pro?fessor in the Depart?ment of Biology and director of Northeastern's Antimi?cro?bial Dis?covery Center.

The stan?dard thinking at the time was that the three main classes of bac?te?ri?cidal antibi?otics each had a unique way of killing bac?te?rial cells -- like spe?cial?ized assas?sins each trained in a single type of weaponry. But this new research sug?gested that all antibi?otics work the same way, by urging bac?te?rial cells to make com?pounds called reac?tive oxygen species, or ROS, which bac?teria are nat?u?rally sus?cep?tible to.

"If they were right it would have been an impor?tant finding that could have changed the way we treat patients," said Iris Keren, a senior sci?en?tist in Lewis' lab.

And that's exactly how sci?ence usu?ally works, said Lewis -- through chal?lenges to main?stream thinking. But recent results reported by Lewis, Keren, and their research part?ners in an article pub?lished Friday in the journal Sci?ence sug?gest that this alter?na?tive hypoth?esis doesn't hold up. For example, even bac?teria that are inca?pable of making ROS are still vul?ner?able to antibi?otics. Fur?ther, some antibi?otics can work their fatal magic in both aer?obic and anaer?obic conditions -- but reac?tive oxygen species can only form when there's oxygen to fuel them.

"We chose to do the sim?plest and most crit?ical exper?i?ment aimed at fal?si?fying this hypoth?esis," said Lewis. "Killing by antibi?otics is unre?lated to ROS pro?duc?tion," the authors wrote. The find?ings were cor?rob?o?rated by Uni?ver?sity of Illi?nois researchers in another study released on Friday .

The team treated bac?te?rial cul?tures with antibi?otics in both the pres?ence and absence of oxygen. Other than the gaseous envi?ron?ment, the two treat?ments were iden?tical. There was no dif?fer?ence in cell death between the two populations.

Before per?forming these exper?i?ments, Lewis' team first looked at sig?nals of a flu?o?res?cent dye, which pre?vious researchers had used as an indi?cator for ROS levels. The team treated bac?te?rial cells with a variety of antibi?otics and mea?sured the strength of this signal. Since antibi?otics were pre?sumed to increase ROS levels, one would have expected increased con?cen?tra?tions of antibi?otics to cor?re?late with stronger sig?nals. How?ever, Lewis' group saw no such correlation.

"But there's a dif?fer?ence between cor?re?la?tion and direct obser?va?tion," Keren said. In order to sup?port their obser?va?tions with unequiv?ocal data, the team mem?bers phys?i?cally sep?a?rated the cells that had stronger flu?o?res?cent sig?nals from those with weak sig?nals and treated them both with the same antibi?otics. Both pop?u?la?tions suf?fered equiv?a?lent cell death.

"The research from Dr. Lewis' group demon?strates that, con?trary to cur?rent dogma, antibi?otics appar?ently do not kill bac?teria through induc?tion of reac?tive oxygen species," said Steven Projan, vice pres?i?dent for research and devel?op?ment at iMed and head of Infec?tious Dis?eases and Vac?cines at Med?Im?mune, both sub?sidiaries of AstraZeneca. "The results shown are rather clear but still leave us with the mys?tery as to how antibac?te?rial drugs help infected people clear bac?te?rial infections. At this point, we should prob?ably dis?pense with the 'one size fits all' approach to bac?te?rial killing by antibi?otics," said Projan, who was not involved in the research.

With these results, Lewis and Keren hope the field will be able to focus its efforts on under?standing the true mech?a?nisms of how antibi?otics wipe out bac?teria in order to effec?tively address chronic bac?te?rial infec?tions, one of the most pressing issues facing public health today.

How the daffodil got its trumpet

Mar. 9, 2013 ? The daffodil is one of the few plants with a 'corona', a crown-like structure also referred to as the 'trumpet'. New research suggests that the corona is not an extension of the petals as previously thought, but is a distinct organ sharing more genetic identity with stamens, the pollen-producing reproductive organs.

The origin of the corona has long been a subject of debate in botany, and in the 1930s botanist Agnes Arber claimed that it was an extension from the petals. With its colourful petal-like appearance, it's easy to see why this was believed for so long. Yet by studying the corona's development and genetic information, this new study has shown that it is in fact related to stamens.

Dr Robert Scotland of the University of Oxford led the research, and was supported by colleagues at Harvard University, the United States Department of Agriculture and the University of Western Australia. The researchers were funded by the Gatsby Charitable Foundation and the United States National Science Foundation. The study is published online in The Plant Journal.

By studying the development of daffodil flowers, the researchers found that the corona only begins to form after the other parts of the flower are fully established. 'This shows that the corona could not be a straightforward modification of either petals or stamens,' explains Dr Scotland, 'Since it develops independently of both, it is more accurately described as a separate organ.'

The different parts of daffodil flowers are located on a small cup-like platform termed the 'hypanthium'.

The researchers analysed genetic activity in all parts of the daffodil flower, and found that daffodil coronas were genetically similar to the stamens and hypanthium, but not the petals.

'We found that the corona develops from the hypanthium, and is not simply en extension of the petals or stamens,' says Dr Scotland, 'The corona is an independent organ, sharing more genetic identity with stamens, and which develops after the other organs are fully established.'

TAMPA, Fla. (AP) ? New York Yankees captain Derek Jeter singled sharply to left field on his first pitch since breaking an ankle last fall in the AL championship series.

Jeter missed New York's first 13 spring training games and was the Yankees' designated hitter and leadoff batter Saturday against Atlanta. Jeter received a partial standing ovation from the crowd at Steinbrenner Field in the first inning, then singled between shortstop and third base against left-hander Mike Minor.

Jeter was forced at second base on Ichiro Suzuki's grounder, sliding into second base on a play that was not close.

The 38-year-old broke his left ankle lunging for a grounder against Detroit last Oct. 13 and had surgery a week later. He returned on the 147th day after the injury and wore protective gear on the foot.

Also, Phil Hughes threw 10 pitches in his first mound session since being diagnosed three weeks ago with a bulging disk in his upper back. The right-hander said he felt great and planned to have a 25-to-30 pitch bullpen session Monday

New York relievers David Robertson (shoulder) and David Aardsma (groin) also had bullpen sessions.

CC Sabathia, the expected opening-day starter, is to pitch in a simulated game Sunday. He had offseason left elbow bone spur surgery.
